Denosumab, marketed by Amgen under the brand names Prolia and Xgeva, is a fully human monoclonal antibody that inhibits RANK ligand (RANKL) to suppress osteoclast formation and bone resorption. Prolia is prescribed for osteoporosis and bone loss associated with hormonal therapy, administered as a 60 mg subcutaneous injection every six months, while Xgeva is indicated for preventing skeletal-related events in patients with bone metastases, giant cell tumors of bone, and hypercalcemia of malignancy, typically given as 120 mg every four weeks following initial loading doses.
